Opis: The three dimensional structure of many extracellular proteins is stabilized by the formation of disulphide bonds. Studies suggest that a microsomal enzyme known as Protein Disulphide Isomerase (PDI) is involved in disulphide-bond formation and isomerization, as well as the reduction of disulphide bonds in proteins. PDI, which catalyses disulphide interchange between thiols and protein dilsulphides, has also been referred to as thiol:protein-disulphide oxidoreductase and as glutathione:insulin transhydrogenase because of its role in reduction of disulphide bonds. The highly conserved sequence Lys-Asp-Glu-Leu (KDEL) is present at the carboxy-terminus of PDI and other soluble endoplasmic reticulum (ER) resident proteins including the 78 and 94 kDa glucose regulated proteins (GRP78 and GRP94 respectively). The presence of carboxy-terminal KDEL appears to be necessary for ER retention and appears to be sufficient to reduce the secretion of proteins from the ER. This retention is reported to be mediated by a KDEL receptor.

Opis: The mammalian protein disulphide-isomerase (PDI) family encompasses several highly divergent proteins involved in the processing and maturation of secretory proteins in the ER by catalyzing the rearrangement of disulphide bonds. PDI, an abundant protein of the ER (>400uM), contains a carboxy-terminal retention signal sequence, KDEL, similar to that of BiP and Grp94. The PDI proteins are characterized by the presence of one or more domains of ~95-110 amino acids related to the cytoplasmic protein thioredoxin. All but the PDI-D subfamily are composed entirely of repeats of such domains, with at least one domain containing - and one domain lacking - a redox-active-Cys-X-X-Cys-tetrapeptide. In addition to roles as redox catalysts and isomerases, PDI proteins perform such functions as peptide binding and cell adhesion, and may conduct chaperone activities. Platelet surface thiols and disulphides play an important role in platelet responses. Catalytically active PDI resides on platelet surfaces where it mediates platelet aggregation and secretion by reducing disulfide bonds, thus exposing fibrinogen receptors in platelets.

Opis: p-Nitroblue Tetrazolium Chloride is an NADPH-diaphorase substrate that competitively inhibits NOS (nitric oxide synthase) (IC50 = 3-4 μM). In combination with 5-Bromo-4-chloro-3-indolyl phosphate (BCIP) and an appropriate reaction buffer, Nitroblue tetrazolium forms a substrate for the colorimetric detection of alkaline phosphatase conjugates in a variety of molecular and cell biology techniques.

p-nitro blue tetrazolium is used for the detection of alkaline phosphatase on western blots. It has also been used as a redox indicator for other enzymatic reactions including dehydrogenases, threonine deaminase, glucose-6-phosphate dehydrogenase, phosphofructokinase on polyacrylamide gels, oxidases on polyacrylamide gels, and pentose shunt dehydrogenases. Redox and halfwave potentials have been determined for p - nitro blue tetrazolium. It has also been used as a colorimetric indicator of bacterial infection in blood samples.
