Opis: The 53-7.3 monoclonal antibody is specific for mouse CD5, a 67 kDa protein that is expressed on most thymocytes, mature T cells and the B-1 subset of B cells. CD5 is a member of the scavenger receptor cysteine-rich protein superfamily and is a ligand for the c-type lectin CD72. CD5 is also known as Leu-1 and functions to modulate antigen receptor signaling on T and B cells.

Opis: The RA3-6B2 antibody reacts with the human and mouse CD45 isoform known as CD45R, or B220, a protein tyrosine phosphatase of 220 kDa. CD45 is one of the most abundant hematopoietic markers, and is expressed on all leukocytes (the Leukocyte Common Antigen, LCA). Various isoforms are generated and expressed in cell-specific patterns, all critical for leukocyte function. In mouse, the CD45R/B220 isoform is predominantly found on B cells, at varying levels on all stages from pro-B cells to activated B cells, and may also be detected on certain T cell and NK cell subsets. It is of note that B220 is not similarly expressed on human B cells, where it appears to be differentiation-specific and therefore expressed on only some B cell subsets. Other forms of CD45 with restricted cellular expression include CD45RA, CD45RB, CD45RO and several others. The RA3-6B2 antibody is one of the most consistently used leukocyte markers for B cells, T cell subsets and NK cell subsets in human and mouse. This antibody has also been reported as cross-reactive with feline CD45R/B220.

Opis: The H57-597 antibody is specific for the beta chain of the mouse T cell Receptor (TCR). This cell surface protein combines with a second protein chain (alpha chain) to form the alpha-beta TCR that is expressed by NK1.1+ thymocytes, NKT cells, and the majority of peripheral T cells. A small number of T cells may express an alternative heteromer of gamma/delta protein chains, known as the g/d TCR. These receptors participate in a complex with CD3, and with the co-receptors CD4 or CD8, to recognize and respond to antigens bound to MHC molecules on antigen-presenting cells. Such interactions promote T cell receptor signaling (T cell activation) and can result in a number of cellular responses including proliferation, differentiation, production of cytokines or activation-induced cell death.</p><p>The H57-597 antibody is used as a phenotypic marker for T cells expressing the alpha-beta TCR. It is also widely used to cross-link surface TCR and thereby mimic TCR-mediated cell activation or induction of apoptosis. The antibody does not cross-react with cells expressing the g/d TCR.

Opis: The PK136 antibody is specific for mouse NK1.1, a type II transmembrane lectin-like receptor and member of the killer cell lectin-like receptor (KLR) family. NK1.1 is prominently expressed on natural killer (NK) cells, and is correlated with NK cytotoxic effects toward virus-infected cells and tumor cells. NK1.1 is expressed on subsets of NKT cells in certain mouse strains (C57BL/6, FVB/N, and NZB), yet absent from others (AKR, BALB/c, CBA/J, C3H, DBA/1, DBA/2, NOD, SJL, and 129). Putative subsets of NK cells and their expression of NK1.1 antigen are of continuing interest, including NK1.1+/CD117+ (c-Kit) cells reported to be immunosuppressive for CD8+ T cells in a mechanism involving PD-1 and PD-L1 (Ehlers et al. 2012. Endocrinology. 10: 1247). The PK136 antibody may be used for detection of NK1.1 expression on mouse strains including CE, B6, NZB, C58, Ma/My, ST, SJL, and FVB. The antibody is reported to react with an epitope common to NKR-P1B and NKR-P1C alloantigenic forms of NK1.1 (Carlyle et al. 2006. J. Immunol. 176: 7511-7524).

L-Selectin is a member of a family of Selectin molecules which act together with the integrin family of adhesion molecules to mediate leukocyte-endothelial interactions. Opis: The GK1.5 antibody reacts with mouse CD4, a 55 kDa protein which acts as a co-receptor for the T cell receptor (TCR) in its interaction with MHC Class II molecules on antigen-presenting cells. The extracellular domain of CD4 binds to the beta-2 domain of MHC Class II, while its cytoplasmic tail provides a binding site for the tyrosine kinase lck, facilitating the signaling cascade that initiates T cell activation. CD4 is typically expressed on thymocytes, certain mature T cell populations such as Th17 and T regulatory (Treg) cells, as well as on dendritic cells.

Opis: The fragmentation of genomic DNA by cellular nucleases during the later stages of apoptosis is also one of the most easily measured features of apoptotic cells. Nuclease activity generates DNA fragments ranging from ~300 bp to 50 bp in length, resulting in a typical DNA ‘laddering’ appearance when analyzed by agarose gel electrophoresis. These fragments have exposed 3’-hydroxyl (OH) ends which can be labeled with bromolated deoxyuridine triphosphates (Br-dUTP). An enzyme, terminal deoxynucleotidyl transferase (TdT), is used to catalyze the template-independent addition of Br-dUTP to the 3’-OH ends of double or single stranded DNA. This method is often called TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) or end labeling. Sites where the Br-dUTP is incorporated can then be detected with an antibody specific to BrdU. With the APO-BrdUTM Kit cells are first labeled with Br-dUTP, and then sites of incorporation are detected through staining with a FITC anti-BrdU antibody. Samples can then be analyzed via flow cytometry. Samples that are apoptotic will stain brightly with the anti-BrdU antibody due to the substantial number of exposed 3’-OH sites, while cells that are non-apoptotic will not have incorporated significant amounts of Br-dUTP and will stain dimly. The APO-BrdU Kit is shipped in one container and consists of two packages. Upon arrival one should be stored at 2-8°C and the other at -20°C.
