Opis: Lipid peroxidation is a well-established mechanism of cellular injury in both plants and animals, and is used as an indicator of oxidative stress in cells and tissues. Lipid peroxides, derived from polyunsaturated fatty acids, are unstable and decompose to form a complex series of compounds. These include reactive aldehydes, of which the most abundant is malondialdehyde (MDA). Therefore, measurement of malondialdehyde is widely used as an indicator of lipid peroxidation. Increased levels of lipid peroxidation products have been associated with a variety of chronic diseases in both humans and model systems.

Opis: The diffusible free radical gas nitric oxide (NO) affects a variety of physiological functions, and is a key regulator of the cardiovascular, nervous, and immune systems. NO is synthesized in many tissues from L-arginine, oxygen, and NADPH by three known isoforms of a heme-containing flavoprotein termed NO synthase (nNOS/NOS-I, iNOS/NOS-II, and eNOS/NOS-III). nNOS is a constitutively expressed neuronal NOS isoform that exits in its latent form until it is activated by the binding of calmodulin follo wing elevation of intracellular calcium levels. The C-terminus of nNOS contains a conserved serine residue, Ser1417, analogous to Ser1177 of the constitutively expressed endothelial NOS isoform (eNOS). Phosphorylation of Ser1417 is believed to regulate nNOS activation, particularly in glucose-sensing neurons, where inhibition of AMPK pathways by glucose and leptin serve to suppress nNOS activity, whereas activation of AMPK by insulin leads to nNOS activation.

Opis: Heme Oxygenase-1 (HO-1) also known as Hsp32, is the inducible isoform of heme oxygenase that catalyzes the NADPH, oxygen, and cytochrome P450 reductase dependent oxidation of heme to carbon monoxide, ferrous iron and biliverdin which is rapidly reduced to bilirubin. These products of the HO reaction have important physiological effects: carbon monoxide is a potent vasodilator and has been implicated to be a physiological regulator of cGMP and vascular tone; biliverdin and its product bilirubin are potent antioxidants; "free" iron increases oxidative stress and regulates the expression of many mRNAs (e.g., DCT-1, ferritin and transferring receptor) by affecting the conformation of iron regulatory protein (IRP)-1 and its binding to iron regulatory elements (IREs) in the 5'- or 3'- UTRs of the mRNAs. To date, three identified heme oxygenase isoforms are part of the HO system that catalyze heme into biliverdin and carbon monoxide. These are inducible HO-1 or Hsp32, constitutive HO-2 that is abundant in the brain and testis, and HO-3 which is related to HO-2 but is the product of a different gene. The HO system is the rate-limiting step in heme degradation and HO activity decreases the levels of heme which is a well known potent catalyst of lipid peroxidation and oxygen radical formation.
